Vitamin B12 (Methylcobalamin)

Methylcobalamin is a naturally occurring cobalamin in which a methyl group occupies the upper axial position on the cobalt centre, in place of the cyano group found in cyanocobalamin. It is produced by microbial fermentation and purified to a dark red crystalline powder. Because effective doses are measured in micrograms, the pure material is usually supplied alongside a 1% trituration on a mannitol or dicalcium phosphate carrier, which makes accurate dry blending practical.

What it is

The cobalamins are corrinoid molecules built around a central cobalt atom, and the group attached above that cobalt distinguishes one form from another. In methylcobalamin it is a methyl group bound directly to the metal through a carbon-cobalt bond. Commercial production runs a selected bacterial fermentation, after which the cobalamin is extracted, converted to the methyl form and crystallised. For the supplement and pharmaceutical trade, a measured portion of that crystalline material is blended with an inert carrier to produce a 1% trituration with verified homogeneity.

Formulation considerations

Light protection is not optional. The carbon-cobalt bond breaks under illumination, so every step from weighing to primary packaging needs to limit exposure, and clear bottles or transparent sachets will shorten shelf life regardless of what the formulation otherwise does. The vitamin is also vulnerable to strong oxidising and reducing agents, which puts ascorbic acid, ferrous salts and sulphites in the category of co-ingredients requiring stability assessment rather than casual co-formulation. The trituration flows and blends predictably, and its pink tint provides a useful visual indicator of distribution during mixing. Dose uniformity should still be confirmed by assay on the finished blend given the inclusion rate involved.

Applications

Methylcobalamin is used in tablets and capsules, sublingual and orally disintegrating tablets, B-complex and multivitamin premixes, oral sprays and drops, and gummies. Available in bulk quantities, with pricing on application.

Handling and storage

Store in sealed original packaging below 15°C where possible, in a dry, dark area, and keep opened containers refrigerated and tightly closed. Typical shelf life is 24 months unopened. Methylcobalamin is markedly light-sensitive and also degraded by oxidising and reducing agents, so weigh under subdued or yellow light, use amber or opaque containers, and protect the finished dosage form from light.

Common questions

What is the difference between methylcobalamin and cyanocobalamin?

They differ in the ligand at the cobalt's upper axial position, a methyl group versus a cyano group. Cyanocobalamin is more stable to light and heat and cheaper; methylcobalamin is the form more often requested for label positioning and requires tighter light protection throughout manufacture.

Should the pure material or a 1% trituration be specified?

At microgram dose levels a trituration is almost always the practical choice, because it can be weighed and blended to uniformity on normal plant equipment. Pure crystalline material suits liquid formulations and operations with suitable micro-dosing capability.

Why does methylcobalamin need protection from light?

The carbon-cobalt bond is photolabile and cleaves on exposure to light, converting the vitamin to inactive degradation products. This drives the use of amber packaging, subdued lighting during handling, and light-protective coatings or opaque capsules in the finished form.

What documentation is supplied with each lot?

Each lot carries a Certificate of Analysis stating assay or declared content, loss on drying, residual solvents and heavy metals, accompanied by the Technical Data Sheet and Safety Data Sheet.
